“In this research, niobium oxide, a solid acid catalyst was evaluated as a pretreatment agent for delignification of KPT-8602 in vivo Alamo switchgrass. The objectives were to determine the effects of temperature, catalyst loading, and pretreatment time on delignification and enzymatic hydrolysis of switchgrass and evaluate reusability of the catalyst. Batch experiments were performed using a Box-Behnken statistical model to study the effects of temperature, selleck inhibitor pretreatment
time, and catalyst loading followed by hydrolysis using Cellic (R) Ctec2 (Novozymes). Niobium oxide was able to reduce total lignin concentrations up to 44.6 +/- 0.97%. Hydrolysis experiments performed for 72 and 168 h (7% enzyme loading) indicated that a maximum glucose yield of 0.169 g g(-1) (59.94% conversion)-0.196 g g(-1) (77.51% conversion) was obtained. Catalyst reusability studies suggested that niobium oxide was able to pretreat four separate batches of switchgrass without losing activity. “Three cyclopeptide alkaloids, nummularine-R (1), nummularin-C (2), and hemsine-A (3), isolated from locally used antidiabetic plant Ziziphus oxyphylla Edgw (Rhamnaceae), showed a potent alpha-glucosidase
inhibition, and moderate anti-glycation activities but no inhibition for alpha-chymotrypsin. This is the first report of the antidiabetic activities of cyclopeptide alkaloids. All compounds Selleckchem Batimastat 1-3 exhibited inhibition for alpha-glucosidase enzyme having IC(50) = 212.1 +/- 1.6 mu M, 215.1 +/- 1.2 mu M, and 394.0 +/- 2.4 mu M respectively, which are more potent than the standard, 1-deoxynojirimycin (IC(50) = 441.0 +/- 0.1 mu M). Compounds 1 (IC(50) = 720.2 +/- 10.9 mu M), and 3 (IC(50) = 277.7 +/- 7.6 mu M) were also found to have a moderate anti-glycation activity. All compounds were non-toxic to PC-3 cell line.
